Washing Lines / Equilibrating Columns

This section washes the selected lines with 40 mls of buffer to purge the system. If you select to equilibrate your column, please check the flow rate. The pressure limit is set to the maximum using this section.

Desalting Runs

HiPrep 26/10 Desalting Column
This section generates a program for a HiPrep 26/10 Desalting Column on an Akta Prime with a Superloop.
Equilibrating the column takes 100ml. Each loop of the column uses 100 ml of buffer.
It will collect 2 x 6.5 ml fractions for each loop, BUT PUT 3 TUBES IN PER LOOP. Sometimes it skips a tube between loops.
Flush Line(s) will flush buffer B if the percent B is not 0. Flushing a pump requires 50 ml of buffer.

HiTrap Desalting Column
This section generates a program for a 5 ml HiTrap Desalting Column on an Akta Prime with a Superloop.
Equilibrating the column takes 20 ml. Each loop of the column uses 15 ml of buffer.
Flush Line(s) will flush buffer B if the percent B is not 0. Flushing a pump requires 50 ml of buffer.

Size Exclusion Runs

XK 16/60 Superdex Columns
This section generates a program for an ~130 ml Superdex Column on an Akta Prime with an optional Superloop.
Equilibrating the column takes 180 ml. Each loop of the column also uses 150 much buffer.
The default collection volumes are designed to collect a peak somewhere between 10 and 20 kDa (RhoGDI) and allow you to run the program 5 times with a full fraction collector.
Flush Line(s) will flush buffer B if the percent B is not 0. Flushing a pump requires 50 ml of buffer.
